On the other hand, the second method—extracting DNA from the remains of the deceased—is considerably more intricate and requires precision,” he explained.

Samples collected from the deceased, like bones or teeth, are meticulously cleaned to remove any external contamination. “If the sample is a bone, it’s ground into powder. If it’s a tooth, it’s broken into small pieces and subsequently powdered. The DNA is then isolated using specialized machines at controlled temperatures,” Sanghvi continued. The extracted DNA undergoes quality and quantity assessment using RT-PCR machines. Only DNA that meets the required quality standards is replicated.

The DNA strands are separated and processed through sequencing machines to create a complete DNA profile. However, Sanghvi warned that if sufficient DNA alleles are not identified, the entire procedure must be restarted. “Identification can only be confirmed when 23 alleles match between the victim and their family members,” he noted.

In cases involving father-son relationships, additional Y-chromosome testing is performed to verify identity. He highlighted the complexity of this work, stating, “Despite the technical hurdles and the time involved, FSL has successfully identified a substantial number of victims in record time with high accuracy.”

In the meantime, the identity of former Gujarat Chief Minister Vijay Rupani has been officially validated through DNA testing, just three days after the tragic crash of Air India flight AI171. This confirmation was made at 11:10 a.m. on Sunday, following exhaustive forensic analysis at the Gandhinagar FSL lab. Rupani's remains are set to be transported from Ahmedabad to Rajkot via a chartered flight.
